<p>L&rsquo;imagerie d&rsquo;une coupe transversale préparée par FIB nécessite à la fois une imagerie de la surface et une imagerie inclinée. Ceci est rendu possible par l’utilisation de divers détecteurs, dont le SED et l&rsquo;UED, permettant d&rsquo;obtenir une imagerie MEB en coupe transversale.</p>

Cette grande platine permet de traiter et d&rsquo;imager la surface entière d&rsquo;un échantillon de 130 mm de diamètre. Les axes de développement se sont concentrés autour de la préparation rapide de lames MET, de l’analyse 3D de la nano-structure de tout type de matériaux et de l’érosion de formes géométriques variables.</h3>
<p>Le JIB-4700F possède une lentille objectif ultra-conique permettant d’atteindre une résolution de 1,6 nm à 1 keV (résolution calculée par séparation de deux particules d’or : ‘gap method’ par opposition à ‘edge method’). Grâce au canon FEG IN-LENS (brevet JEOL), des courants très importants (300 nA avec une image à la clé !) dans une petite taille de sonde permettent d’explorer les limites spatiales de L’EDS et de l’EBSD en 3D. La nouvelle colonne FIB à ions gallium permet de focaliser une sonde, avec un courant allant jusqu’à 90 nA, pour les procédés ultra-rapide de fabrication ionique de piliers ou de lames minces.</p>
<p>Le dernier axe de développement est la robustesse de ce FIB. En effet, son design a été optimisé pour garantir une stabilité et une fiabilité accrues lors des procédés FIB &amp; MEB.</p>

<p style="text-align: justify;">La conception unique de la cartouche CRYO ARM<sup>TM</sup> assure un maintien optimal de la grille MET, qui conserve toujours la même orientation durant l’ensemble du processus. Ce dispositif simplifie non seulement le workflow de préparation, mais également celui de la microscopie corrélative (CLEM), permettant le transfert des coordonnées des zones d’intérêt entre les différents instruments.</p>
<p style="text-align: justify;">Les cartouches CRYO ARM<img src="https://s.w.org/images/core/emoji/15.0.3/72x72/2122.png" alt="™" class="wp-smiley" style="height: 1em; max-height: 1em;" /> présentent une polyvalence appréciable dans le processus de préparation d’échantillons pour la cryo-MET. Elles permettent en effet d’accueillir une coupelle de 3 mm, obtenue par la méthode <em>High Pressure Freezing</em> (HPF). Grâce à cette configuration, il est possible de réaliser la préparation de lames minces par <em>lift-out</em> ou d’effectuer une acquisition en Volum EM. Cette flexibilité contribue à optimiser le workflow, en s’adaptant aux différentes méthodes de préparation et d’analyse des échantillons vitrifiés.</p>

Pour le refroidissement et le maintien en température de la platine cryo, il est important de limiter les vibrations induites par le système de refroidissement (phénomène présent pour les systèmes refroidis à l’azote liquide). C’est pour cela que JEOL a intégré un refroidissement par conduction thermique sur la platine cryo qui réduit ainsi la dérive et les vibrations de cette dernière. Ce développement améliore grandement la stabilité et préserve l’état vitrifié de l’échantillon. Cette solution améliore la résolution qui est la même à température ambiante et en mode cryo.</p>

Sa conception innovante permet de limiter efficacement la formation de glace, un problème fréquent pouvant altérer la qualité des échantillons.<br />
Grâce à ce système, il est désormais possible de traiter et de préparer de grandes quantités d’échantillons tout en maintenant un niveau de propreté optimal dans la chambre. Cette intégration réduit le nombre de transferts et limite grandement les risques de contamination ou de dévitrification de l’échantillon. Ce workflow est compatible avec tous les microscopes électroniques en transmission, qu’ils soient équipés d’un autoloader cryo, d’un porte-objet classique ou qu’ils fonctionnent avec le système AutoGrid.</p>

Cette configuration permet de conserver une orientation spécifiquement adaptée à l’acquisition tomographique. Grâce à cette orientation, la collecte des données est facilitée et le nombre d’acquisitions possibles est augmenté. En effet, la découpe de la surface d’intérêt s’effectue perpendiculairement à l’axe d’inclinaison du porte-objet, ce qui optimise l’accès à la région d’étude.</p>
