<p>Il existe 2 familles de 3D en microscope électronique :</p>
<ul>
<li>La première passe par la création de stacks d’images en 2D en cross section sur MEB ou le FIB (array tomography, « SBF » Serial Block Face ou FIB 3D).</li>
<li>La seconde passe par le calcul des projections images de l’échantillon tilté obtenues en MET (tomographie).</li>

L’imagerie de chaque section (ou de zones d’intérêts sur chaque section) est faite avec un MEB, le volume reconstruit final est l’assemblage du stack d’images. L’Array Tomography est une technique non destructive, les coupes obtenues par ultramicrotomie sont déposées sur wafer ou lame de verre avec dépôt conducteur et sont conservées.</p>

En répétant l’opération automatiquement, on obtient une série d’images 2D du bloc. L’assemblage des images du stack nous permet d’obtenir la visualisation 3D complète du bloc. Des extractions ultérieures de zones (segmentations) ou d’objets d’intérêts (cellules, organelles, etc.) permettent de révéler les structures 3D d’objets µmétriques. Coupes et observations sont enchainées automatiquement grâce aux logiciels dédiés. Les coupes échantillons étant « sacrifiées » , cette technique est destructive.</p>

En fin d’acquisition une reconstruction 3D est obtenue par assemblage du stack d’images 2D. Les échantillons seront au préalable enrobés et contrastés avec des métaux lourds selon différents protocoles. Cette technique à plusieurs avantages : elle permet une grande précision de positionnement de la découpe, présente des distorsions plus faibles et une meilleure résolution 3D (sur le plan en Z due à l’épaisseur des coupes =20nm). Cette technique permet aussi de s’affranchir des inconvénients liés à la découpe mécanique et permet de découper tous types d’échantillons : durs, fragiles, cassants, comme des dents, os, métaux, silicium, etc. Les coupes échantillons étant « sacrifiées » , cette technique est destructive.</p>

Les reconstructions par calculs (qui utilisent les images des projections de l’échantillon tilté en continu) permettent de visualiser en 3D des structures intracellulaires, protéines ou organelles de très petites tailles en très haute résolution.</p>

Cela permet d’éviter l’absorption de rayons X par le matériau de la fenêtre, il est donc possible d’analyser des éléments légers tels que le béryllium (Be) et le bore (B). Par conséquent, le détecteur EDS sans fenêtre a été davantage utilisé ces dernières années.</p>

<p>Elle fournit des données précises et quantitatives sur la morphologie et la cristallographie d&rsquo;un matériau, par exemple la forme, la taille et les relations de voisinage des cristaux, les phases cristallographiques, la texture cristallographique et la distribution des désorientations. Outre la description morphologique des microstructures, cette technique permet la caractérisation cristallographique complète des joints de grains et la quantification des structures de déformation en termes de densités de dislocations géométriques nécessaires.<br />
L&rsquo;EBSD fournit une image avec une résolution spatiale de ～0,1 μm et une résolution angulaire de ～1°. La plage angulaire acquise par EBSD est de ～20°. Normalement, l&rsquo;EBSD est réalisée dans un MEB, mais elle peut également être réalisée dans un MET.</p>
<p>Expérimentalement, l&rsquo;EBSD est réalisée avec un microscope électronique à balayage (MEB) équipé d&rsquo;un détecteur EBSD comportant au moins un écran phosphorescent, un objectif compact et une caméra CCD ou CMOS. Il est nécessaire de s’équiper d’un bon détecteur mais aussi d’un microscope électronique à balayage ayant des capacités de courant très élevé pour gagner en temps, efficacité et également explorer des nouvelles conditions (par exemple à basse tension). Il est également possible en combinant le mode LDF (<em>large depth of focus</em>) du <a href="https://www.jeol.fr/produit/jsm-it810/" target="_blank" rel="noopener">JSM-IT810</a> de faire des cartographies grand champ sans déformation optique et sans bouger la platine.</p>

Le temps d’illumination est un indice de la vitesse de balayage du faisceau d&rsquo;électrons.<br />
Lorsqu’il est multiplié par le nombre de pixels de balayage dans un balayage de ligne horizontale, puis que le temps de retour en arrière est ajouté, on obtient le temps de balayage pour un balayage de ligne horizontale. Lorsque ce temps de balayage est multiplié par le nombre de balayages verticaux (lignes de balayage horizontales), le temps d&rsquo;acquisition d&rsquo;une image de balayage est calculé.</p>
